The incubation temperature should be optimal for the activity of the antibody (i.e., 4(C or 37(C), and the incubation time can vary, usually from one to several hours. The usual volume of serum-to-cells is one volume serum to one volume of washed packed red cells. To adsorb high titre antibodies, it may be necessary to use several aliquots of the same cells and do multiple adsorptions in order to remove all of the antibody.
