Method

  1. A master dilution is prepared by doing a two-fold (doubling) dilution (1:2, 1:4, 1:8, etc.) of the patient's serum in a diluent. The diluent can be saline, bovine albumin (usually 6%) or "inert" AB serum.

  2. Drops from each master dilution are transferred to separate tubes, appropriate red cells are added, and an appropriate technique is done for the antibody in question (e.g., saline 4°C, albumin 37°C, IAT, etc.)

    NOTE: The larger the volume of the master dilution, the greater the accuracy of the titration. Also, once master dilutions are made, the titration should be done as soon as possible (antibodies are unstable in the diluted state).
