Sanger Sequencing
Effective November 13, 2020 TAGC will no longer be offering Sanger Sequencing services. November 12th will be our last run, as such the last day for sample drop off will be Tuesday November 10th by 4:00pm. As of November 13, 2020 Sanger Sequencing can be done at the Department of Biological Sciences Molecular Biology Facility, MBSU.
Genotyping
TAGC offers customized protocols for DNA extraction and PCR amplification from mouse and rat tissues (e.g. ear and tail) using the KAPA Mouse Genotyping Kit. The PCR products are examined on the QIAxcel, a fully automated capillary electrophoresis system, which reports both gel images and electropherograms, and determines the size and concentration of DNA fragments.
Nucleic Acids Evaluation
Determination of the size, concentration and quality of DNA and RNA is performed on automated electrophoresis instruments, which significantly increase the number of samples that can be processed at a time and produce a normalized digital output that can be used for comparisons within a run or between various runs. TAGC currently has two automated DNA/RNA Electrophoresis instruments and one dye-based nucleic acid quantification instrument:
Qiagen QIAxcel
Agilent Bioanalyzer 2100
Qubit Fluorometer
These instruments are available to be run by TAGC technicians as a service at a rate of $80/hr + reagent costs.
Real-Time PCR
TAGC houses a 7900HT Fast Real-Time PCR System, with 3 interchangeable blocks, for:
96-well fast plates
96-well standard plates
384-well fast plates
The type of assays that can be performed on the 7900HT are:
- Allelic discrimination (quantitative analysis)
- Relative quantification of gene expression
- Absolute quantification
- Dissociation curve
The 7900HT supports both Taqman and SYBR green assays.
TAGC provides training for $80. Upon completion of this training users can book and run the instrument during TAGC hours at a cost of $20/run.
